| PA1-781 detects mouse and rat melanopsin. PA1-781 has been successfully used in Western blot, immunofluorescence, and immunocytochemistry procedures. By Western blot, this antibody detects ~53 and ~85 kDa proteins representing overexpressed unglycosylated and glycosylated melanopsin from HEK 293 cells (does not work well on endogenously expressed protein). By immunofluorescence and immunocytochemistry, PA1-781 detects melanopsin from rat retinal ganglion cells (RGCs). The PA1-781 immunogen is a synthetic peptide corresponding to residues E(455) Q K S K T P K T K R H L P S L D R R M(474) of rat melanopsin. Figure 1 shows immunofluorescence staining of melanopsin in rat retinal ganglion cells using PA1-781. |
| General Information |
| Melanopsin is an opsin-like protein found in the retinal ganglion cells of mammals. Melanopsin is believed to be part of a secondary optical system that parallels that of the classic rod and cone system. This second system reports directly to the suprachiasmatic nucleus and is responsible for regulation of circadian rhythms. Melanopsin is believed to be the primary photopigment responsible for the regulation of these circadian rhythms, and Melanopsin knockout mice have been generated which demonstrate decreased capacity to entrain to light and dark cycles. |
