PA1-1751 detects ADAMTS5 from porcine, bovine, and human samples. PA1-1751 is specific for ADAMTS5 and shows no cross-reactivity with other ADAMTS family members.
PA1-1751 has been successfully used in Western blot procedures. By Western blot, this antibody detects a major band at ~68 kDa protein representing ADAMTS5 from porcine cartilage extract. On human recombinant ADAMTS5 this antibody detects a major band at ~85 kDa. Depending on the extent of C-terminal processing, several smaller bands may also be seen using this antibody.
PA1-1751 immunogen is a synthetic peptide corresponding to residues C K(636) N G Y Q S D A K G V K T F(649) of human ADAMTS5. This polyclonal antibody has been referred to as: JSCKNG.
Figure 1 shows a Western blot of ADAMTS5 in porcine cartilage extract demonstrating a major band at ~68 kDa and a second band at ~50 kDa using PA1-1751. Figure 2 shows a Western blot of ADAMTS5 in human recombinant protein demonstrating a major band at ~85 kDa and a second band at ~68 kDa using PA1-1751. |
A disintegrin-like and metalloproteinase thrombospondin type 1 motif 5 (ADAMTS5 or aggrecanase 2) is a member of the ADAMTS family of multidomain proteins that has demonstrated metalloproteinase functions. The ADAMTS5 protein contains a prohormone convertase cleavage site, thrombospondin-1 like domain, a zinc-binding motif, a disintegrin-like domain, cysteine-rich domain, and a propeptide domain. ADAMTS5 expression is found in a wide range of human tissues, including the placenta, bladder, rib cartilage, cervix and joint tissue.
ADAMTS5 plays an important role in the cleavage of aggrecan and studies have determined that aggrecan is cleaved within the interglobular domain of aggrecan at site Glu(373)-Ala(374). Aggrecan degradation has been linked to the development of arthritic diseases such as rheumatoid arthritis and osteoarthritis as well as retinal disease. |
