| OPA1-01011 detects the caspases-8 (FLICE (FADD (fas-associated via death domain) -like ICE) ) inhibitory protein (FLIP) from human cells. This antibody shows no cross-reactivity with the short form of FLIP, FLIPS. OPA1-01011 has been successfully used in Western blot procedures. By Western blot, this antibody detects a single ~55 kDa protein representing the long form of FLIP, FLIPL, in HeLa cell lysate. OPA1-01011 immunizing peptide corresponds to amino acid residues 447-464 from human FLIP. OPA1-01011 immunizing peptide (Cat. # OPEP-01011) is available for use in neutralization and control experiments. Figure 1 shows a Western blot of FLIP in HeLa cell lysate using OPA1-01011. |
| General Information |
| Apoptosis is related to many diseases and induced by a family of cell death receptors and their ligands. Cell death signals are transduced by death domain (DD)-containing adapter molecules and members of the ICE (interleukin-1-beta converting enzyme)/CED-3 (C. elegans death receptor-3) protease family. Caspases-8 (FLICE (FADD (fas-associated via death domain) -like ICE)) and -10 (FLICE2 (FADD (fas-associated via death domain) -like ICE 2)) are two pivotal members in the ICE/CED-3 protease family. FLICE-inhibitory proteins (FLIPs) were identified in virus and humans and designated v-FLIPs and FLIP, respectively. FLIP contains two death effector domains (DEDs) and a caspase-like domain. FLIP interacts with adapter protein ADD and caspase-8 and -10, and potently inhibits apoptosis induced by all known death receptors DR3 (death receptor 3), TRAIL-R (TNF-related apoptosis-inducing ligand receptor) and TNFR1 (tumor necrosis factor receptor 1). |
